Ham’s F-12 (F-12) was designed for serum-free, single-cell plating of Chinese Hamster Ovary (CHO) cells. Compared to other basal media, Ham’s F-12 contains a wider variety of components such as zinc, putrescine, hypoxanthine and thymidine. Ham’s F-12 has been used for serum-free growth of CHO cultures as well as serum-supplemented growth of other cells, including chondrocytes and rat prostate epithelial cells.

Ham’s F-12 contains no proteins or growth factors. Therefore, Ham’s F-12 requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). Ham’s F-12 uses a sodium bicarbonate buffer system (1.18 g/L), and therefore requires a 5–10% CO₂environment to maintain physiological pH.

All raw material components used for Servicebio cell culture media products are screened through using strict quality control testing. Servicebio cell culture media are manufactured at a cGMP-compliant facility using water for injection, filtered through final 0.1 μm sterile filtration and aseptic filled in a Hundred-stage FFU. The filter integrity tests are performed before and after filtration. Each lot of cell culture media are subjected to sterility test, pH, osmolality, endotoxin and cell culture test.

The major character of this Ham’s F-12 is as follows:

l With 10 mM Glucose, 1.0 mM L-glutamine, 1.0 mM sodium pyruvate, phenol red.

l Without HEPES.
